The solute carrier organic anion transporter family member 1B1 (SLCO1B1) gene encodes a membrane-bound sodium-independent organic anion transporter (OATP1B1) that is involved in the active transport of many endogenous and xenobiotic compounds. The SLCO1B1 gene is located on chromosome 12p12.2 and has fifteen exons. Although many variants have been identified in SLCO1B1, the decreased function variant c.521T>C (p.V174A; rs4149056) has been implicated in reduced simvastatin pharmacokinetics and is associated with an increased risk for statin-induced muscle toxicity. The c.521T>C variant is found in the SLCO1B1*5, *15, and *17 haplotypes.
This targeted genotyping test interrogates the following variants: c.521T>C (p.V174A; rs4149056).
- Anticoagulated blood: Two 5-10 mL tubes with EDTA (lavender top) are preferred. ACD tubes (yellow top) are also accepted
